Radioimmunoassay for dexamethasone in plasma.

چکیده
Until recently there has been no adequate clinical method for precisely measuring dexamethasone in blood. Studies of therapeutic dexamethasone concentrations and of its metabolism in plasma have relied on the use of radioactive dexamethasone (1-3). We report here an immunoassay in which antisera obtained from rabbits immunized with a conjugate of dexamethasone and bovine serum albumin (BSA) are used. The assay measures plasma concentrations of dexamethasone down to 0.5 izg/liter.

Plasma corticotropin radioimmunoassay, with use of fuller's earth for extraction.
A radioimmunoassay is described for plasma corticotropin whereby concentrationsover the range of 6 to 1000 ng/liter can be measured. Corticotropin extracted from plasma on fuller’s earth under carefully standardized conditions is assayed by a long-used double antibody radioimmunoassay procedure. Within (n = 6) and between (n = 12) assay precision is 39 ± 2 (SD) ng/liter and 44 ± 6 (SD) ng/liter...

We report the utilization of a very sensitive radioimmunoassay for plasma B-END, which has enabled us to examine the suppressibility of B-END-like material, under dexamethasone challenge, on a population of depressed patient~. We have found that B-END-like material in plasma is suppressed by dexamethasone in a psychiatric control population, whereas suppression is less likely in endogenously de...
متن کاملLiquid chromatography and radioimmunoassay compared for determination of cortisol and corticosterone in plasma after a dexamethasone suppression test.
We developed sensitive, specific "high-performance" liquid chromatography (HPLC) for determining suppressed cortisol and corticosterone in human plasma and compared its efficacy with that of conventional radioimmunoassay (RIA) at concentrations in the nanogram per liter range. Steroids from a 0.5-mL aliquot of plasma were extracted by rapid-flow fractionation, with diethyl ether as mobile-phase...
متن کاملUse of the Dexamethasone-Corticotrophin Releasing Hormone Test to Assess Hypothalamic-Pituitary-Adrenal Axis Function in Rheumatoid Arthritis
Objectives. Hypothalamic-Pituitary-Adrenal axis function may be abnormal in rheumatoid arthritis (RA). A pilot study in 7 patients suggested impaired glucocorticoid feedback in some patients after the dexamethasone-corticotrophin releasing hormone (CRH) test. This study aimed to investigate the dexamethasone-corticotrophin releasing factor test in a larger group of patients and relate the resul...
متن کاملComparison of plasma cortisol and corticosterone in the dexamethasone suppression test for melancholia.
The suppression of plasma corticosterone (B), measured by radioimmunoassay (RIA), was compared to simultaneous suppression of plasma cortisol (F), measured as total corticoids by a competitive protein binding (CPB) assay, in the overnight dexamethasone suppression test (DST). Baseline plasma B concentrations in 10 control subjects were 4.04 +/- 1.07 ng/ml (X +/- S.D.) at 0800 hr and 1.51 +/- 0....
